| 摘 要： | 对红酵母细胞壁的分离纯化及细胞壁主要成分的含量检测方法进行了分析研究。制备红酵母细胞壁需要经过破壁、色素提取、除杂等过程。酵母细胞壁的主要成分是葡聚糖、甘露糖、蛋白质和几丁质。碱法水解是从酵母细胞壁中提取检测β-1,3-D-葡聚糖的理想途径。费林试剂络合法提纯分离红酵母细胞壁多糖,可得到纯化的甘露聚糖。几丁质含量的检测一般通过间接测定其水解产物N-乙酰氨基葡萄糖的含量来实现。通过定氮仪测定氮元素含量可以转化测定蛋白质的含量。

| Abstract: | The isolation and detection of the content for rhodotorula cell wall were studied.The preparation of rhodotorula cell wall was composed of breaking cell wall,extracting pigment and purification.Glucan,mannan,protain and chitin were the main components of rhodotorula cell wall.Alkali method was the perfect method for extracting and examing β-1,3-D-glucan from rhodotorula cell wall.The mannan could be extracted and purified by FehLing reagent method from rhodotorula cell wall.Examation of chitin according to the method of examining the content of N-acetylglucosamine.The protein content was calculated indirectly by determining the nitrogen content of cell wall in a nitrogen analyzer. |
