摘    要:为给林下栽培平菇资源的综合利用提供参考依据,采用Box-Behnken响应面法优化了以超声辅助酶法提取富硒平菇柄多糖的工艺条件,并测定了多糖硒含量。结果表明,最佳提取条件为:复合酶液(纤维素酶∶果胶酶=2∶3)用量为2.5倍样品量,酶解温度47℃,超声提取时间46 min。在此工艺条件下,富硒平菇柄多糖的提取得率为14.53%,其硒含量为10.12μg/g。验证实验结果表明,此提取工艺简单可行,且按此工艺条件制得的富硒平菇柄多糖中有机硒的含量丰富,具有较高营养价值。

Abstract:In order to provide some references for comprehensive utilization of Pleurotus ostreatus resources planted in forest,the ultrasound assisted enzymatic extraction conditions of polysaccharides from selenium enriched P ostreatus were optimized by using Box-Behnken response surface methodology.The selenium content in polysaccharide was also determined.The results showed that the optimal extraction conditions were as follows:the amount of composite enzymes (cellulase ∶ pectinase =2;3) was as 2.5 times as the sample,enzymolysis temperature was 47 ℃C,ultrasonic time was 46 min.Under the conditions,the extraction rate of polysaccharides was 14.53%,and selenium content was 10.12 μtg/g.The results of verification test showed that the conditions were simple and feasible,selenium enriched P ostreatus had rich selenium content in stem and had higher nutrient value.
