大鼠β微管蛋白在大肠杆菌中的表达及其抗体制备

摘    要:在获得重组质粒pMD-18T—BTUB的基础上,成功地构建了重组表达质粒pET-28a—BTUB,并将其转化入大肠杆菌BL21(DE3)感受态细胞中,经IPTG诱导表达,该诱导菌经SDS上样缓冲液进行处理后,SDS—PAGE电泳检测到相对分子质量约为14400的目的蛋白带,薄层扫描分析表明目的蛋白占总蛋白的41.8%。目的蛋白经透析袋电洗脱法纯化后,采用常规方法制备家兔多克隆抗体,ELISA检测表明此蛋白具有良好的抗原性。

Abstract:On the base of recombinant plasmid pMD 18T BTUB,the recombinant expression plasmid pET 28a BTUB was constructed successfully.Then the recombinant was transformed into E.coli stain BL21(DE3),which induced by IPTG when D 600 of the culture was about 0 6.The special expression protein(about 14 400 ) was detected by SDS PAGE.The expression protein amounts to 41 8% in the total mass of bacterial protein by thin layer scan.After purified by electroelution into dialysis bags,the special expression protein was used to make polyclonal antibody by routine method,ELISA detection showed the protein have a satisfactory antigenicity.
