温度对家蚕微孢子虫(Nosema bombycis)孢子体外发芽的影响

摘    要:【目的】调查温度对家蚕微孢子虫(Nosema bombycis)孢子体外发芽的影响,为家蚕微粒子病的防治及其他微孢子虫的基础研究提供参考依据。【方法】以K2CO3-KHCO3法诱导孢子体外发芽,在孢子发芽过程中利用不同温度(5、15、25和35℃)在不同时期处理家蚕微孢子虫孢子,观察不同处理的孢子发芽率。【结果】在前期4个温度处理的家蚕微孢子虫孢子发芽率分别为3.88%、15.66%、31.91%和58.75%,孢子发芽率随着温度的升高而增大;在刺激期4个温度处理的孢子发芽率依次为53.33%、38.17%、25.07%和6.08%,孢子发芽率随着温度的升高而减小;在培育期4个温度处理的孢子发芽率分别是44.63%、41.29%、43.53%和48.82%,无显著差异。【结论】温度对家蚕微孢子虫孢子体外发芽具有一定的影响,在家蚕微孢子虫孢子体外发芽过程中,前期低温和刺激期高温能抑制孢子发芽,而前期高温和刺激期低温能促进孢子发芽。

Abstract:Objective The present study was conducted to investigate the effect of temperature on the germination of Nosema bombycis spore in vitro so as to provide information for preventing and treating pebrine and relevant microsporidia fundamental research. Method The spores were stimulated to germinate with the way of K2CO3-KHCO3 at different temperature (5℃,15℃,25℃, 35℃) at different period, then germination rate from different treatment was observed. ResultIn earlier stage, the germination rate of Nosema bombycis spores was 3.88%, 15.66%, 31.91% and 58.75%, respectively. The germination rate of Nosema bombycis spores went up with the rising temperature. The germination rate of Nosema bombycis spores was 53.33%, 38.17%, 25.07% and 6.08% in stimulated stage, respectively, and it went down with the rising temperature. The ger-mination rate of Nosema bombycis spores was 44.63%, 41.29%, 43.53% and 48.82% in rearing stage, respectively, and there was no significant difference. Conclusion Temperature had certain effect on germination of Nosema bombycis spores in vitro. Throughout the entire process of spore in vitro germination, low temperature in earlier stage and high temperature in stimulated stage could inhibit the germination of Nosema bombycis spores, and high temperature in earlier stage and low tem-perature in stimulated stage could promote its germination.
