摘    要:【目的】验证紫外分光光度法测定葛粉工业废水葛根素含量的可靠性,建立一种可信度高、操作性强、适于监测葛粉工业废水葛根素含量动态变化的测定方法。【方法】在不同浓度的葛根素标准溶液中加入已知浓度的牛血清蛋白(BSA),检验蛋白质对紫外分光光度法测定葛根素含量的干扰;分别采用紫外分光光度法和高效液相色谱(HPLC)法测定葛粉工业废水葛根素含量,确认紫外分光光度法测定废水中葛根素含量的可靠性。【结果】在2~20μg/mL浓度范围内,葛根素浓度与其在250nm处的吸光值呈良好的线性关系,线性方程为:y=0.0648x-0.0049,相关系数(R2)为0.9995。紫外分光光度法测定葛根素的最低检出限为0.0480μg/mL,具有精密度较高、准确度良好,且不受BSA干扰的特点。紫外分光光度法与HPLC法测定结果的相对误差为5.0%,在可接受范围内。【结论】利用紫外分光光度法替代HPLC法进行葛粉工业废水葛根素含量测定具有可信、简便的特点,适用于工厂中葛粉工业废水葛根素分离纯化生产过程的实时监测。

Abstract:Objective The reliability of content determination of puerarin in radix puerariae starch industrial wastewater by UV method was verified to select a reliable, operative, and suitable method for observing content variation of puerarin in the wastewater. Method Puerarin standard solutions with various concentrations were mixed with bovine serum albumin (BSA) to test the interference of protein on UV method. To confirm the reliability of determining puerarin content in wastewater using UV method, the results of UV and HPLC methods were compared. ResultThe calibration curve of puerarin showed a satisfying linear relativity in the range of 2-20 μg/mL with R 2 =0.9995.(Linear equation: y=0.0648x-0.0049). The detection limit of UV method with no BSA interference was 0.0480 μg/mL,which showed high precision and accuracy. The relative error between the results of UV and HPLC methods was 5.0%, which belonged to the acceptable range. Conclusion The determination of puerarin in radix puerariae starch industrial wastewater by UV method instead of HPLC was easy to operate and the result was accurate. Therefore, UV method could be used to the real time supervision of the separation of puerarin from radix puerariae starch industrial wastewater.
