两系杂交稻亲本SSR指纹图谱的建立及其在种子纯度鉴定中的应用

摘    要:利用筛选到的20对SSR引物建立了两系杂交稻32个亲本(24个光温敏不育系和8个恢复系)的DNA指纹图谱,针对所涉及的9个杂交稻组合,获得能在父、母本间表现出多态性的特异SSR标记48个.以杂交稻组合两优932为例,在实验室用一个特异SSR标记(RM302)对200粒种子样本进行了纯度鉴定,结果鉴定纯度为90.50%,与田间种植鉴定结果90.60%(海南鉴定)和91.57%(武汉鉴定)基本一致,表明SSR标记技术适用于构建DNA指纹图谱和种子纯度鉴定.

Abstract:In this research, DNA fingerprints of 24 male sterile lines and 8 restorer lines of two-line hybrid rice were constructed by using 20 SSR (Simple sequence repeats) primers. The 20 SSR primers could amplify 70 polymorphic bands totally with a mean of 3.5 bands per primer. 48 specific SSR markers for identifying the seed purity of 9 two-line hybrid rice combinations were acquired.The 200 single seeds of Liangyou 932 were tested by a specific SSR marker (RM302) in laboratory. The seed purity was 90.50%, which was approximate to the field purity (90.60% in Hainan and 91.57% in Wuhan, respectively). The results of this research suggested that SSR technique could be used in DNA fingerprinting construction and seed purity identification of two-line hybrid rice.
