四倍体青贮玉米染色体加倍及检测技术研究

摘    要:以辽单527、辽单青贮178、辽原一号3个玉米品种为试验材料,通过6个浓度的秋水仙素溶液处理,进行染色体加倍,采用流式细胞仪检测加倍效果。结果表明,当秋水仙素浓度达0.1%时,3个品种加倍率均达到最高值,辽单527的加倍效果最好,最高可达36.3%。利用流式细胞仪能够快速准确检测染色体倍性,OTTO法制备悬浮液的检测效果较好。

Abstract:In order to detect the double effect by flow cytometry(FCM), there varieties of silage maize including Liaodan527, Liaodan178 and Liaoyuan No.1 were chosen as the materials, and 6 concentrations of colchicine were used for chromosome doubling. The results showed that, the doubling rate were all reached the highest level when the concentration of colchicine was 0.1%, and that of Liaodan527 was the best, which was 36.3%; FCM could correctly detect the chromosome multiple and OTTO method had better effect.
