五角枫初代培养影响因素研究

摘    要:以五角枫幼嫩茎段作为外植体,对五角枫初代培养的灭菌时间、外植体采集时期、基本培养基、植物生长调节剂组合等进行了研究。结果表明:采用0.1%HgCb灭菌剂对五角枫茎段外植体进行灭菌,最佳灭菌时间为4 rnin;4月至5月是五角枫茎段外植体较为理想的采集时期,1月至3月是五角枫当年生枝条经水培后茎段外植体的最佳采集时期;MS培养基是五角枫初代培养的最佳基本培养基;培养基添加1.0 mg/L 6-BA,0.2 mg/L NAA与0.5 AC时,腋芽诱导率最高,达92.4%;五角枫初代培养的最佳pH值为5.8.

Abstract:The disinfection time,collection period of explants,types of basic culture medium and the combination of plant growth regulators of primary culture of Acer mono were studied with the infancy stem segments of Acer mono were used as the explants.The results showed that sterilization with 0.1%HgCl2 for 4 min was the best sterilization time for stem explants of Acer mono.The better collection period for explants was from April to May,and April to May was the best collection period for infancy stem segments of current-year branches after hydroponics.The optimum medium was MS medium for primary culture.The induction rate of axillary bud was the highest(92.4%)when MS medium supplemented with 1.0 mg/L BA,0.2 mg/L NAA and 0.5 g/L AC.The optimum pH was 5.8 for primary culture.
