罗田甜柿幼胚培养条件的研究

摘    要:利用 5因素 4水平的正交试验对‘罗田甜柿’幼胚培养条件进行了探讨 ,结果表明 :(1)培养基以MS(1/ 2N)最适 ;(2 )蔗糖浓度以 7%为宜 ;(3)抗褐化剂以活性炭效果最好 ;(4 )最佳激素配比是IBA 0 .3mg/L +Zeatin0 .44mg/L ;(5 )激素种类生长素类以IAA为好 ,细胞分裂素类以Zeatin效果较好 ;(6 )MS(1/ 2N) +IAA 1mg/L +暗处理 4d生根率最高

Abstract:The factors influencing immature embryo culture in Luotiantianshi persimmon variety ( Diospyros kaki f.) were studied by Orthogonal design in 5 levels and 4 planes. The results showed that: (1) the optimum medium was MS(1/2N); (2) the optimum sucrose concentration was 7%; (3) activated carbon was the best for browning control; (4) the optimum IBA and Zeatin concentration was 0.3 mg/L and 0.44 mg/L ,respectively; (5) the optimum kind of auxin and cytokinin was IAA and Zeatin; (6) a higher frequency of rooting occurred on half strengthen MS(1/2N) medium supplemented with 1 mg/L IAA and dark treatment for 4 days.
