| 摘 要： | 利用ArrayDesigner2.0软件设计PCV2引物直接从PDNS病料肺组织中PCR扩增获得PCVCQB7基因片段,用pMD18-T载体连接、克隆获得PCVCQB7重组质粒。序列测定和与标准毒株对位比对分析PCVCQB7核苷酸序列全长624bp,系PCV2ORF2基因片段,与PCV2分离株的核苷酸序列相似性在80%以上,与PCV1在66%以下,采用最大似然法构建的系统发生树分析发现PCVCQB7位于PCV2主枝上,并与国内和欧洲分离株Imp.1147、304、375、QD、GD-TS、SH、24657_NL毒株亲缘关系较近形成一小分枝,表明PCVCQB7为引起PDNS的PCV2扩增产物。

| Abstract: | PCVCQB7 was amplified from lung samples porcine dermatitis and nephropathy syndrome(PDNS)directly by PCR and sequenced.It is located in ORF2 of PCV type 2(PCV2)and is 624bp length.By comparison of PCVCQB7 and PCV,its nucleotides identities are above 80%(PCV2)and below 66%(PCV1),while its nucleotides and amino acids identities are above 94% and 90% with isolates from PDNS.Phylogenetic tree based on PCVCQB7 nucleotide sequence by Maximum likelihood shows that the PCVCQB7 is located in PCV2 major branch and forms a minor branch with Imp.1147,304,375,24657_NL,QD,GD-TS,SH isolates from European and China.It is concluded that PCVCQb7 is ORF2 segment of PCV2 caused PDNS. |
