摘    要:本研究利用Wx基因微卫星特异性分子标记"484/485"对13份国内常用保持系进行PCR检测,筛选出具有中等AC含量、Ⅰ型带型的优质保持系"宜香1B"作为优质供体.配制"协青早B/宜香1B",对其F_1、F_2代单株用"484/485"PCR分子检测,证明其带型符合一对基因分离模式.在此基础上,对‘协青早B/宜香1B"的杂交、回交和自交的低世代群体(BE_1F_1、BC_1F_2)进行"484/485"分子标记辅助选择,再结合优质不育系其它性状选择,快速育成了中等AC含量、垩白率低、透明度高、不育特性和农艺性状稳定的籼型优质三系小育系"浙农3A",并于2009年9月顺利通过了浙江省品种审定委员会组织的专家现场鉴定.本文还就利用"484/485"分子标记有效改良稻米AC含量等问题进行了讨论.

Abstract:In the research,genotypes of the 13 indica maintainer lines were detected by the Wx gene microsatellite molecular marker"484/485",and the result showed that Yixiang 1B had the type Ⅰ Wx gene allele,which had a moderate apparent amylose content(AC).This microsatellite was then used to genotype the F_1 and F_2 individuals derived from the cross Xieqingzao B/Yixiang 1B,and found that the Wx marker followed a one-ene segregation pattern.Screening the individuals of the BC_1F_1 and BC_1F_2 derived from further cross,backcross and self-cross of Xieqingzao B/Yixiang 1B with this microsatellite marker were carded out.Finally,a good quality CMS line Zhenong 3A were bred,which showed stable and complete male sterility,good agronomic characters,low degree of chalkiness,high degree of transparency,and intermediate AC.Zhenong 3A was registered by the Zhejiang Provincial Crop Variety Appraisal Committee in 2009.How to efficiently improve the AC of rice grain using Wx marker was also discussed in this paper.
