丹顶鹤源弗氏柠檬酸杆菌的分离鉴定及药敏结果分析

摘    要:目的 为查明一例源于安徽省动物园的丹顶鹤死因。方法 从该丹顶鹤肝部组织坏死灶采集病料,进行细菌分离与鉴定,并对其药敏结果进行分析。结果 该菌落在血琼脂平板上生长呈白色不透明菌落,革兰氏阴性菌;生理生化试验显示该菌在葡萄糖、甘露醇、乳糖、山梨醇和硫化氢等中均呈阳性;PCR 扩增其 16S rDNA 基因,其大小约 1500bp,经遗传进化树分析,与已报道的弗氏柠檬酸杆菌NBRC 12681株(NR 114345.1)在同一分支;药敏结果显示其对氧氟沙星和庆大霉素高度敏感,对丁胺卡那、多粘霉素B中敏;对青霉素、阿莫西林、四环素、头孢噻肟、氨苄西林、复方新诺明、氟苯尼考耐药。结论 本次从患病丹顶鹤肺部分离的病原是弗氏柠檬酸杆菌,且对多种抗菌药具有一定的耐药性。为珍稀野生保护动物的相关病原研究和防治提供参考。

Isolation and Identification of Cithombacterium Freundi from Red-crowned Crane and Analysis of Drug Susceptibility Results
Abstract:Objective to identify the cause of death of red-crowned crane from zoo in Anhui Province. Methods The liver tissue necrosis of the crane was collected from the disease material, the bacteria were isolated and identified, and the drug susceptibility results were analyzed. Results The colony grew on blood agar plates as white opaque colonies and gram-negative bacteria. Physiological and biochemical tests showed that the bacteria were positive in glucose, mannitol, lactose, sorbitol and hydrogen sulfide. PCR amplifies its 16S rDNA gene, which is about 1500 bp in size, and is analyzed by genetic evolutionary tree in the same clade as the reported NBRC 12681 strain of Citrobacter Freundi (NR 114345.1); The drug susceptibility results showed that it was highly sensitive to ofloxacin and gentamicin, and sensitive to bumicana and polymyxomycin B. Resistant to penicillin, amoxicillin, tetracycline, cefotaxime, ampicillin, co-trimoxazole, and florfenicol. Conclusion The pathogen isolated from the lungs of the diseased red-crowned crane is Citherobacterium Freundi and has a certain resistance to a variety of antibacterial drugs. It provides reference for the research and prevention of pathogens related to rare wild protected animals.
