摘    要:本文详细分析了水稻广亲和中粳品系02428胚性悬浮细胞系建立过程中的培养液渗透值、PH值,呼吸作用,过氧化物酶同工酶,碳、氢及矿质元素的变化。结果如下:1、前期、中期和后期悬浮细胞系(好胚性悬浮细胞系)的培养液渗透值在一次继代培养的0-3天期和后期悬浮细胞比前期悬浮细胞更多地利用氨基酸作为代谢底物。后期悬浮细胞的EMP途径相对稍低,而HMP、TCAC途径较前期、中期稍强。3、从前期-中期-后期,过

Abstract:The authors analysed the changes of the osmotic potential, pH values, respiration, isoperoxidase and the contents of carbon, hydrogen and mineral elements in the culture solutions during the establishment of embryogenic suspension cell lines of rice (cv. 02428). The results are as follows: 1. The osmotic potential in the culture solutions of the suspension cell lines at the former phase (FP), middle phase(MP) and later phase (LP) increased rapidly 0-3 day after subculture and reached the peak in 5 days. The change of pH value was just the opposite. 2. Suspension cell lines at MP and LP utilized more amino acid for metabolic substrate than that at FP. The EMP pathway level in suspension cell line at LP was relatively lower but the HMP and TCAC levels higher than in those at FP and MP. 3. From FP to MP to LP, the isoperoxidase activity in the cathode region decreased significantly. The isoperoxidase activity in the anode region of suspension cell line at LP was also lower than that at FP. 4. Compared with suspension cell line at FP and MP, the nitrogen and magnesium contents in suspension cell line at LP were higher while calcium and iron contents were lower. The above-mentioned physiological and biochemical changes are somewhat related to whether the suspension cell lines are suitable for protoplast culture.
