滇重楼的花粉活力测定方法比较

摘    要:目的]为提高滇重楼的繁殖效率。方法]将滇重楼开花植株裂开花药的花粉轻轻涂抹载玻片上,分别用碘-碘化钾(I2!KI)染色法、氯化三苯四氮唑(TTC)染色法、醋酸洋红染色法和略有改动的荧光素二醋酸酯法对滇重楼花粉活力进行测定。结果]氯化三苯基四氮唑染色法的染色率较低,仅为27.51%;醋酸洋红染色法和荧光素二醋酸酯法的染色率较高,分别为91.30%和90.55%,而且所测数据较为接近;碘-碘化钾染色法虽然能将花粉染上色,染色率为94.80%,但染上色的花粉包含退化的花粉和失活的花粉。结论]醋酸洋红染色法和荧光素二醋酸酯染色法的染色率较高,能较好地区分出有活力的花粉和失活的花粉,而且能较为准确地反映滇重楼花粉的生活力。

Abstract:Objective The study aimed to increase the reproduction efficiency of Paris polyphylla var.yunnanensis.Method The pollens of dehisced anthors of flowering P.polyphylla plants was slightly painted on the slide glass,the activities of P.polyphylla pollens were determined resp.by iodine-potassium iodide dyeing method(I2-KI),triphenyl tetrazolium chloride staining method(TTC),aceto carmine dyeing method and fluorescein diacetate method with a little improvement.Result The dyeing rate of TTC method was lower,being only 27.51 %;that of aceto carmine dyeing method and fluorescein diacetate method were higher,being 91.30 % and 90.55 % resp.,and the determined data were close to each other.Although I2-KI method could dye pollens with dyeing rate of 94.80 %,the dyed pollens included degraded and inactive ones.Conclusion Aceto carmine dyeing method and fluorescein diacetate method has higher dyeing rate.The active and inactive pollens can be distinguished better and the viability of P.polyphylla pollens can be reflected more accurately.
