摘    要:番茄废渣经微生物发酵可制成蛋白饲料。本试验以番茄废渣为原料,研究3,5-二硝基水杨酸(DNS)法测定番茄废渣及其发酵产物中还原糖及总糖含量的最优方案。结果表明,测定波长为520 nm,DNS试剂用量为1.5 mL,显色时间为5 min,盐酸浓度为6 mol/L,控制在20 min内完成测定。此方法测得的结果精密度高、重现性好,平均回收率可达99.85%,RSD为0.671%,表明采用DNS法测定番茄废渣中的糖分含量准确、可行。

Study on Determination of Sugar Content from Tomato Residue with 3, 5-Dinitrosalicylic Acid Colorimetry

Abstract:Tomato residue could be made into protein feed through microbial fermentation. The method of DNS was applied in this article in order to analyze the content of reducing sugar and total sugar in tomato residue and its fermentation product. The result showed that, the optimal conditions as follows, wave lengh was 520 nm, the volume of DNS solution was 1.5 mL, reaction time was 5 minutes, concentration of HCl was 6 mol/L and ensuring complete determination within 20 minutes. The results of the experiment had the characteristics of high precision, good repeatability, the average recovery rate was 99.85%, and RSD was 0.671%. It was showed that using DNS method to determine the sugar content of tomato residue was accurate and feasible.
