摘    要:以小麦"5389"的幼胚为外植体诱导愈伤组织,研究了培养基中2,4-D、KT、ABA及其配比对愈伤组织诱导和愈伤组织继代培养的影响。结果表明:以MS+2,4-D 3.0 mg/L培养基的诱导效果最好,出愈率最高达93.42%;在MS+2,4-D 1.0 mg/L+KT 0.1 mg/L培养基上进行继代培养获得松脆型愈伤组织,以MS+2,4-D1.0 mg/L+KT 0.1 mg/L培养基进行液体培养建立了状态良好的悬浮培养细胞。

Abstract:Rataria of wheat 5389 were cultured to study the effect of different mediums on callus induction and the estate of the callus.The results showed that in MS+2,4-D 3.0 mg/L culture medium the highest ratio of callus induced was 93.42%,and in MS+2,4-D 1.0 mg/L+KT 0.1 mg/L maintenance medium the embryonal callus were obtained and cell suspension culture of wheat 5389 was established.These results provided a good system for further study on how the elicitor can induce the defense reaction of host.
