晚熟甜橙芽变的ISSR鉴定

摘    要:应用ISSR技术对晚熟甜橙伏令夏橙的大果无核枝、"安顺一号"等5个材料进行了鉴定。从100个引物中筛选出18个引物,共扩增出113条带,其中多态性条带数为15条,多态位点百分率为13.3%。结果表明:812、834、841引物对参试材料有明显扩增带差异,伏令夏橙大果无核枝与对照枝及伏令夏橙、"安顺一号"与伏令夏橙及锦橙在DNA分子水平上存在明显的遗传差异,晚熟甜橙大果无核枝为伏令夏橙一稳定芽变。ISSR技术能有效地鉴定晚熟甜橙芽变。

Abstract:DNA polymorphism of C.sinensis cv.were determined by inter simple sequence repeat(ISSR) molecular markers.113 bands were amplified with 18 of 100 primers,among which 15 were polymorphic between mutants and parental cultivars,and the polymorphism ratio was 13.3%.These results showed that there were genetic diversity with primer 812,834 and 841 between seedless big fruit branch and the contrast branch,among 'Anshun No.1' and Valencia and 'Jincheng' orange.Combining the results of morphological and molecular analysis,it was certified that the branch of C.sinensis cv.and 'Anshun No.1' occurred genetic variation.
