茶树花药愈伤组织诱导及增殖研究

摘    要:以茶树花药为外植体,以MS培养基为基本培养基,探究适合花药愈伤组织诱导的花药发育时期,不同光照时间对花药愈伤组织诱导的影响,不同植物生长调节物质对花药愈伤组织诱导及增殖的影响,为建立茶树组织培养体系提供依据。研究结果表明:利于花药愈伤组织的形成的处于单核中期的花蕾直径在0.7~0.8 cm,黑暗条件下较12 h/d光照条件下茶树愈伤组织的诱导率高,茶树花药经过低温预处理24 h后,在培养基MS+6-BA0.5 mg/L+NAA0.4 mg/L+2,4-D1.0 mg/L上愈伤组织诱导率最高,诱导率为84.6%,茶树花药愈伤组织增殖率最高的培养基为MS+NAA1.0 mg/L+6-BA0.5 mg/L+2,4-D0.2 mg/L。

Abstract:With tea tree anther as explants, MS medium as basic medium, we explored the suitable anther development period for anther callus induction, the effects of the different illumination time on anther callus induction, and the effects of different plant growth regulating substances on anther callus induction and proliferation, so as to provide the basis for establishing tea plant tissue culture system. The results showed that the diameter of flower buds at the middle stage of mononuclear was 0.7-0.8cm, which was conducive to the formation of anther callus. The induction rate of tea tree callus under dark condition was higher than that under 12h/d light condition. After low temperature pretreatment for 24h, The highest callus induction rate was 84.6% on MS+ 6-BA 0.5mg/L+NAA0.4mg/L+2, 4-d 1.0mg/L, and the highest proliferation rate of tea anther callus was MS+NAA1.0mg/L+ 6-BA 0.5mg/L+2, 4-d 0.2mg/L.
