摘 要: | 目的]研究紫杉二烯合成酶基因转化短叶红豆杉(Taxus brevifolia)内生真菌XT5的情况。方法]采用土壤农杆菌介导法将来自红豆杉中的紫杉二烯合成酶基因转入红豆杉内生真菌XT5中,研究紫杉醇发酵产量。结果]成功转化得到8株重组子,其中XT5-TS-2的紫杉醇含量得到了大幅提高,其最高产量为423.983 4μg/L,比原始菌株提高了53.19%,表明转化紫杉二烯合成酶基因能够有效提高紫杉醇产量。结论]该研究可为提高红豆杉内生真菌发酵生产紫杉醇产量提供理论支持。

Abstract: | Objective] The aim was to study the transformation of endophytic fungus XT5 by taxadiene synthase gene in T.brevifolia.Method] The taxadiene synthase gene in T.brevifolia was transferred into the endophytic fungus XT5 by soil agrobacterium-mediated method,and then the fermentation yield of taxol was studied.Result] 8 strains of recombinants were successfully transformed,and the toxol yield of XT5-TS-2 improved tremendously with the highest of 423.983 4 μg/L,increasing by 53.19%,which indicated that the transformation of the taxadiene synthase gene could effectively improve the toxol yield.Conclusion] The study could provide theoretical support for improving the toxol yield by the fermentation of endophytic fungus in T.brevifolia. |
