3株乳酸片球菌的鉴定及其耐酸耐盐特性和抑菌作用研究

摘    要:为了探索从日本处理有机废弃物样品中分离的乳酸片球菌(Pediococcus acidilactici)耐酸耐盐特性和抑菌作用,采用浸注培养法从处理有机废弃物样品中进行菌株分离,并对分离菌株R-4、B-7、D-13进行生理生化及16S rDNA鉴定;分别将其接种于pH4的MRS肉汤培养基和50 mg/mL NaCl的MRS肉汤培养基中37℃培养48 h,观察其生长情况;通过牛津杯法测其培养液对大肠埃希菌K88、枯草芽胞杆菌、金黄色葡萄球菌、沙门菌的抑菌作用。结果从日本处理有机废弃物样品中分离得到13株乳酸球菌,其中菌株R-4、B-7、D-13经鉴定均为乳酸片球菌;并在50 mg/mL NaCl和pH4的MRS肉汤培养基中生长良好且呈现较好的耐酸耐盐特性;3株乳酸片球菌的培养液对4种指示菌均呈现抑制作用,其中菌株B-7的培养液对金黄色葡萄球菌的抑菌作用最强,抑菌圈直径达17.02 mm±0.33 mm,但其培养液中酸性产物及H2O2产物排除结果说明,对金黄色葡萄球菌的抑菌作用可能与其在生长过程中产生的酸有关。研究结果为乳酸片球菌特性及其应用的进一步研究提供了科学依据,为筛选出处理有机废弃物的微生物发酵菌奠定了基础。

Abstract:In order to explore the acid-salt tolerance and bacteriostatic action of Pediococcus acidilactici which isolated from Japanese organic waste treatment samples,strains were isolated from organic waste treatment samples by the method of infusion culture.The strains R-4,B-7 and D-13 were identified by physiological-biochemical and 16S rDNA levels;The isolated strains were inoculated in MRS liquid medium of pH4 and 50 mg/mL NaCl at 37°C for 48 h to observe growth characteristics.Bacteriostatic actions of its metabolites against Escherichia coli K88,Bacillus subtilis,Staphylococcus aureus and Salmonella were determined by the Oxford Cup method.The results showed 13 strains of Lactococcus lactis were isolated from samples,and strains R-4,B-7 and D-13 were identified as Pediococcus acidilactici,showed good acid-salt tolerance in MRS liquid medium of 50 mg/mL NaCl and pH4.The three strains showed good bacteriostatic action,strain B-7 showed the strongest bacteriostatic action against Staphylococcus aureus,and the inhibition zone diameter was 17.02±0.33 mm.Exclusion results of culture medium indicated that the inhibition against Staphylococcus aureus may be related to the acid produced during growth process.This study provided a scientific basis for further research on the characteristics and application of Pediococcus acidilactici,and for selecting microbial fermenting strains to organic wastes.
