湖南省部分地区中华竹鼠毕氏肠微孢子虫基因分型研究

摘    要:为了明确湖南省部分地区中华竹鼠毕氏肠微孢子虫的感染情况,从湖南省4个市的6个养殖场共采集中华竹鼠新鲜粪便样品480份,利用小亚基核糖体RNA(SSU rRNA)基因的套式PCR对这些样品中毕氏肠微孢子虫的感染率和遗传多样性进行研究。结果发现,25份粪便样品存在毕氏肠微孢子虫感染,总感染率为5.2%,除吉首市的中华竹鼠未检出阳性外,其余各采样点及各年龄段的中华竹鼠均有毕氏肠微孢子虫感染,但感染率差异不显著(P>0.05);进一步序列分析发现,所检测的中华竹鼠毕氏肠微孢子虫存在2种SSU rRNA基因型(Type 1和Type 2),其中Type 1为优势基因型;多位点序列分析发现,在MS1、MS3、MS7基因位点分别有7、1、14份样品成功扩增,呈现7、1、4个单倍型。本研究结果将为湖南省部分地区中华竹鼠微孢子虫病的防控提供基础数据。

Abstract:To clarify the infection status of Enterocytozoon bieneusi in Rhizomys sinensis in partial areas of Hunan province,a total of 480 fresh fecal samples of R.sinensis were collected from 6 farms in 4 cities of Hunan province and examined for the prevalence and genetic diversity of E.bieneusi in these rodents by using the Nested-PCR targeting the small-subunit ribosomal RNA(SSU rRNA)gene.The results showed the presence of E.bieneusi infection in 25 fecal samples,with the total prevalence of 5.2%.Except that no infections were detected in Jishou city,the infection was found in all other age groups and sampling sites,but the differences in prevalence among them were not significant(P>0.05).Further sequence analysis revealed two genotypes(Type 1 and Type 2)in these rodents,with the Type 1 as the predominant genotype.MLST analysis found that 7,1 and 14 samples were successfully amplified at the MS1,MS3 and MS7 loci,presenting 7,1 and 4 haplotypes.The findings in the present study will provide basic data for the prevention and control of microsporidiosis in R.sinensis in partial areas of Hunan province.
