大豆体细胞胚对卡那霉素和PPT敏感性的研究

摘    要:以大豆(Glycine max(L.)Merrill)品种合丰25、东农L13、东农40、黑农40、黑农35、CH21141、Progress的体细胞胚为材料,采用MS培养基附加2,4-D(15 mg/L~20 mg/L),以不同浓度的卡那霉素和PPT处理.结果表明:大豆对卡那霉素和PPT敏感性在基因型间存在显著差异;随着卡那霉素和PPT浓度的增加,大豆体细胞胚褐化率增加,体积增殖减小;以卡那霉素作为筛选剂,最初使用浓度为50~100 mg/L,而以PPT作为筛选剂,最初使用浓度为2 mg/L.

Abstract:The sensitivity of somatic embryo tissue treated with kanmycinand PPT was studied.The results showed that there was certain difference between the sensitivity of soybeancultivars to selective agents;with the concentrations of kanmycin and PPT increased,the brown frequency of somatic embryo tissue increased,but the proliferation of somatic embryo tissue decreased;the suitable concentrations were kanmycin 100mg/L,PPT 2 mg/L.
