| 摘 要： | 基于Light Cycler平台,利用SYBR GreenI染料能特异与双链DNA结合而发荧光的特性,建立了一种荧光PCR方法检测禽网状内皮组织增殖病病毒(REV)的方法。整个检测过程2 h内可以完成,病毒的最低检出浓度为10-7,在特异性试验中,REV有特异性的扩增曲线,而禽流感病毒和阴性对照一样,没有特异性扩增。对REV患鸡各器官组织进行了荧光PCR检测,各脏器病毒含量从高到低依次是肝、脾、腺胃、肾、肌胃,肝脏中的病毒含量最高,肌胃中的病毒含量最少,肺中没有检测到病毒。基于SYBR GreenI的荧光PCR方法检测REV快速、敏感、特异性好,为开展REV病原学检测提供了一种快速、无污染的方法。

| Abstract: | Based on Light Cycler,using the conjunct speciality of SYBR GreenI dye and double-chain DNA,a detection method of REV using Real-time PCR.All detection course need 2h.The virus minimum detection concentration is 107.In the differential test,REV but AIV is detected.In the detection of sample,the viral content in liver is the highest,but no virus was detected in the lung.It would be a fast and no-pollution method for the pathogeny detection of REV. |
