甘肃省当归种质资源的RAPD分析

摘    要:用筛选出的7个随机引物对来自甘肃省不同产区的20个当归样品的基因组DNA进行了RAPD及其聚类分析,共扩增出82个位点,其中多态性位点58个,占70.7%.聚类分析结果表明:20个当归样品可以聚为5类,其结果与当归的地理分布比较吻合,即甘肃省当归栽培群之间亲缘关系的远近与栽培地的地理分布距离有一定的相关性.地理分布距离较近的样品遗传距离较近,但在同一分布区域内,生态条件对当归遗传差异亦有影响.

Abstract:The genomic DNA of 20 samples of Angelica sinensis(Oliv.) Diels from different producing area in Gansu Province was analyzed by Random Amplified Polymorphism DNA(RAPD) and cluster analysis with 7 selected primers.In the test 82 amplified bands were obtained,among which 58 bands were polymorphic,accounting for 70.7 %.The result showed that all the samples could be clustered into five groups.The clustering result was coincided with the geographic distribution pattern of A.sinensis,that is to say there were some correlations between the genetic relationship among different cultivated populations of A.sinensis and the geographic distance to each other.The genetic distance of different samples was shorter when the geographic distance of two samples was nearer,but even in the same region,different ecological condition also influenced the genetic difference of A.sinensis.
