| 摘 要： | 为了研究家蚕核型多角体病毒(Bombyx mori nucleopolyhedro virus,BmNPV)中GP41蛋白的包装功能,从BmNPV基因组中克隆了gp41基因,并将其克隆到杆状病毒转移载体pFastBac1-gfp中,构建重组转移载体pFastBac1-gp41-gfp,再利用杆状病毒表达系统(Bac-to-Bac)筛选重组杆状病毒,在家蚕BmN细胞系中进行融合表达和定位分析。SDS-PAGE和Western blot检测结果显示,经重组杆状病毒r-gp41-gfp感染的BmN细胞新增一条大小为65 kD左右的蛋白条带,证明该融合蛋白在BmN细胞中成功表达。用激光共聚焦显微镜观察到绿色荧光充满于细胞质中,不能形成聚集体。与野生型BmNPV混合感染的实验也表明荧光出现的位置与多角体无关,说明GP41-GFP蛋白不能附着在多角体表面,融合表达可能影响了GP41蛋白与多角体的结合。
