摘    要:[目的]优化紫花苜蓿下胚轴的再生条件。[方法]选用不同的培养基和激素,对公农1号、公农2号、美国苜蓿王、草原1号4个苜蓿品种的下胚轴进行愈伤组织的诱导和分化。[结果]经筛选,获得了诱导愈伤组织的基本培养基为改良SH+MS有机+MS微量及铁盐+2,4-D2.0mg/L+KT0.05mg/L+蔗糖30g/L,诱导再生植株分化的培养基为MS+NAA1.0mg/L+KT0.1mg/L+水解酪蛋白25mg/L+蔗糖20g/L。[结论]得到公农1号、公农2号、草原1号3个易于诱导和分化的基因型。

Optimization of the Regeneration Conditions from Alfalfa Hypocotyls

Abstract:[Objective] The research aimed to optimize the regeneration conditions of hypocotyls alfalfa.[Method] Different media and hormones were used to induce and differentiate from the hypocotyls on Gongnong 1, Gongnong 2, American Alfaking and Caoyuan 1.[Result] After screening, the basic medium for inducing the callus was obtained as: improved SH+MS organic fractions +MS micronutrients + 2.0 mg/L 2,4-D + 0.05 mg/L KT+ 30 g/L sucrose.The medium for inducing the differentiation of regenerated plants was obtained as MS+1.0 mg/L NAA +0.1 mg/L KT+25 mg/L casein hydrolysate and 20 g/L sucrose.[Conclusion] Three genotypes for the induction and differentiation were obtained from Gongnong 1, Gongnong 2, American Alfaking and Caoyuan 1.
