摘    要:目的]研究旨在分析三江黄牛Prdm9锌指域序列特征。方法]实验选用三江黄牛(n=14),通过PCR扩增其Prdm9基因锌指域后进行克隆测序。结果]结果显示,三江黄牛Prdm9锌指域中存在14种锌指,它们共构成9种锌指域(等位基因),其中1种锌指和4种锌指域在牛属动物上未见报道。各锌指间有1~5个氨基酸的差异,发生在6个位点,其中5个为正向选择位点。结论]三江黄牛Prdm9锌指域的有效等位基因数、基因杂合度、多态性高,具有丰富的遗传多样性。并且通过Prdm9锌指域的分析比较,从一定层面上提示了三江黄牛可能与瘤牛间存在亲缘关系。

Abstract:Objective] The purpose of this study was to analyze the sequence characteristics of Prdm9 zinc finger domain in Sanjiang yellow cattle. Methods]Sanjiang yellow cattle (n = 14) were selected for the experiment. The zinc finger domain of Prdm9 gene was amplified by PCR and then cloned and sequenced. Results]The results showed that there were 14 kinds of zinc fingers in Prdm9 zinc finger domain of Sanjiang yellow cattle, which constituted 9 kinds of zinc finger domains (alleles), of which 1 kind of zinc finger and 4 kinds of zinc finger domains were not reported in cattle. There were 1 ~ 5 amino acid differences among the zinc fingers, which occurred at 6 sites, of which 5 were positive selection sites. Conclusion]The number of effective alleles, gene heterozygosity and polymorphism of Prdm9 zinc finger domain in Sanjiang yellow cattle are high, and there is rich genetic diversity. And through the analysis and comparison of Prdm9 zinc finger domain, it suggests that there may be a kinship between Sanjiang yellow cattle and rumu cattle.
