藏药短穗兔耳草中微量元素的形态分析

摘    要:[目的]建立常用的藏药短穗兔耳草的水煎液和人工胃液中Ca、Fe、Mg、Mn、Zn、Cu、Pb和As8种微量元素的形态分析方法。[方法]利用HNO3-HClO4湿法消解样品,采用0.45μm微孔滤膜分离水煎液中的可溶态和悬浮态微量元素;利用D101大孔吸附树脂柱分离有机态和无机态微量元素;同时制备了样品的人工胃液提取液。采用ICP-OES法依次对原生药及各种形态中的8种元素含量进行测定。[结果]短穗兔耳草中8种元素在原生药、可溶态和人工胃液提取液中的形态分布基本一致。水溶液的提取率在1.48%~44.87%,人工胃液中的溶出率在2.45%~62.82%。方法的加标回收率在92.7%~104.5%,相对标准偏差小于2.48%。[结论]该研究建立的方法可同时快速、简便、准确地测定短穗兔耳草中各形态的8种微量元素,且短穗兔耳草中含有丰富的人体必需微量元素Ca、Mg、Fe、Zn和Mn,可能是其发挥药效的重要原因之一。

Speciation Analysis on Trace Elements in Lagotis brachystachya Maxim

Abstract:[Objective] To develop methods for speciation analysis of trace elements in aqueous solution and simulated gastric fluid of Lagotis brachystachya Maxim,a kind of Tibetan medicines.[Method] Samples were digested by HNO3-HClO4.The water-soluble state and suspension state were separated through microporous filtering film.Water-soluble state was divided into organic state and inorganic state with macroporous adsorption resin.Artificial gastric juice was prepared for extracting trace elements from the samples.The elements were detected by ICP-OES.[Result] The species distribution of the eight trace elements was consistent in the original sample,water-soluble state and gastric juice.The extraction rate of elements was 1.48%-44.87% in the aqueous solution and 2.45%-62.82% in the simulated gastric fluid.The recovery rate of standard addition was 92.7%-104.5%,and the relative standard deviations(RSD)were less than 2.48%.[Conclusion] The developed method is simple,accurate,rapid and convenient Lagotis brachystachya Maxim is rich in essential trace elements including Ca,Mg,Fe,Zn and Mn,which may be important for drug efficacy.
