摘    要:PCR—SSCP是一种新的分子遗传标记方法,但影响其实验效果的因素很多。本研究主要综合有关文献报道,应用3种不同组成的聚丙烯酰胺凝胶(丙烯酰胺/N,N′—亚甲基双丙烯酰胺分别为49:1,39:1,29:1)进行比较分析,其中以聚丙烯酰胺凝胶浓度为8%、丙烯酰胺/N,N′—亚甲基双丙烯酰胺比例为49:1、加5%甘油的组成的效果最佳,其带纹清晰,容易识别。

Abstract:PCR-SSCP is one new kind of methods of molecular markers. There are a lot of factors affecting the result of PCR-SSCP. In this experiment, 3 groups of different components of polyacrylamide gel were used to select the best one for further PCR-SSCP analysis. The results showed that best one is 8% polyacrylamide gel (Acr: Bis, 49:1) with 5% glycerol which can produce clear bands.
