基于单克隆抗体的酶联免疫法与高效液相色谱法检测水产品中环丙沙星残留的比较

摘    要:为了快速、灵敏地检测禁用药物环丙沙星在水产品中的残留,建立了基于单克隆抗体的间接竞争酶联免疫法(competitive indirect ELISA,ci-ELISA),并比较了其与高效液相色谱法(High Performance LiquidChromatography,HPLC)在灵敏度、准确性和重现性等方法学参数上的差异。实验结果显示,经过浓缩、净化等前处理过程,ci-ELISA与HPLC的检测灵敏度分别可以达到10μg/kg和1μg/kg,回收率分别为79.28%~92.92%和86.27%~97.32%,相对标准偏差分别在3.21%~8.88%和2.66%~5.52%。前者经济、高效,适合作为初筛方法大批量筛选样品;后者灵敏度、重现性更好,适于在实验室条件下作为验证方法。
