小黑麦高分子量谷蛋白研究

摘    要:对183份小黑麦进行了高分子量谷蛋白的十二烷基硫酸钠—聚丙烯酰胺凝胶电泳(SDS-PAGE)分析,发现6种新的亚基或亚基组合,暂命名为5*、6*、7*、8*、10*、21*+12*;共检测到38种高分子量谷蛋白亚基组合类型,其中,亚基组合null、7*+8*、6*+20出现频率最高,有53份材料具有该亚基组合。供试小黑麦在Glu-A1位点,null为优势亚基,出现频率为42.62%,优质亚基1和2*出现频率分别为21.86%、28.42%;在Glu-B1位点,20亚基出现频率最高,为56.28%,其次为14+15和13+16亚基,频率分别是17.49%和15.85%,在供试小黑麦中有29份材料具有小麦品种中比较少见的优质亚基13+16。供试小黑麦高分子量谷蛋白在Glu-A1、Glu-B1位点的亚基分布和硬粒小麦非常相近,初步确定供试小黑麦的四倍体亲本可能为硬粒小麦。

Abstract:Six new subunits or subunit combinations named temporarily 5*,6*,7*,8*,10* and 21* 12* were detected from 183 Triticale materials introduced from CIMMYT with SDS-PAGE analysis.There were 38 high molecular weight glutenin subunit combinations detected among 183 Triticale materials,among which,null,7* 8* and 6* 20 were the most frequent combination and there were 53 materials having this kind of combination.Null was an advantageous subunit,which emerging frequency was 42.62 %,and the frequency of good subunits 1 and 2* was relatively high,which was respectively 21.86 % and 28.42 % at locus Glu-A1 of the tested Triticale materials; the subunit 20 appeared most frequently at locus Glu-B1,which frequency was 56.28 %.And then the frequency of subunit 14 15 and subunit 13 16 were respectively 17.49 % and 15.85 %.The good subunits 13 16 was rare in twenty-nine tested Triticale materials.The distribution of high molecular weight glutenin subunits (HMW-GS) of the Triticale materials was very similar to that of T. durum,so we preliminarily concluded that the tetraploid parent of the Triticale materials may be T. durum.
