梅花品种‘淡丰后’茎段开放式启动培养的初步研究

开放式组织培养在植物组织培养的研究发展中,受到了重视。该文作者对梅花品种‘淡丰后’进行了启动培养的初步研究。通过不同基本培养基、激素种类及浓度的对比实验,得出‘淡丰后’以当年生嫩枝作外植体,MS培养基优于WPM培养基;IBA较NAA更能促进芽的萌发;6-BA浓度为2·0mg/L较4·0mg/L更能促进芽的萌发,最终筛选出‘淡丰后’带腋芽嫩茎段的最佳启动培养基为MS+6-BA2·0mg/L+IBA0·5mg/L,并对外植体消毒时间和叶片去留对污染的影响作了初步研究。

Preliminary studies on the open initiation culture of Prunus mume ‘Dan Fenghou’

Researchers always pay attention to open culture. The authors studied the open initiation culture of Prunus mume ‘Dan Fenghou’. The experiments focused on basal culture media, hormone types and concentration. The results showed that it was suitable to use one-year-old twigs as explants. MS culture medium is better than WPM culture medium;IBA is better than NAA to promote sprouting. The 6-BA at 2.0 mg L in the medium showed higher explant sprouting rate than that at 4.0 mg L. The results indicated that the optimal initiation culture medium is MS+6-BA 2.0 mg L+IBA 0.5 mg L. Comparative experiments on disinfection time and blade removal were also made.
