单细胞凝胶电泳检测除虫脲残留对小鼠肝脏DNA损伤的研究

摘    要:除虫脲是一种低毒、低残留、没有致突和致癌性的农药,但是它光解后在蔬菜上的某些残留物,如对氯苯、对氯苯基脲等有一定的毒性。采用单细胞凝胶电泳试验,研究除虫脲在蔬菜上残留的混合物对小鼠肝脏细胞DNA的损伤作用,应用CASP软件分析彗星图像,得出彗星尾部DNA百分含量、彗星尾长、尾距和Olive尾距4个指标,通过SPSS软件分析表明,残留混合物导致的彗星拖尾现象与空白组有显著区别。研究表明,尽管除虫脲原体没有致突性,但是它在蔬菜上残留的混合物具有一定的致突、致畸、致癌性,所以不仅应对其原体化合物进行评价,还必须对它的分解物和代谢物同时进行安全评价。

Abstract:Diflubenzuron,a kind of pesticides,can not induce mutation and aberration because of its low toxicity and low residue.But it's the photolytic residues after photolysis,such as 4-chlorobenzene and chlorophenyl urea,in vegetables might have some toxicity.Single cell gel electrophoresis(SCGE) was performed to study the DNA damage of mice liver cell caused by the mixture of diflubenzuron residual in vegetables.And the Comet Assay Software Project(CASP) software was used to analyze the comet images.Based on four parameters including percentage of tail DNA(TDNA%),tail length(TL),tail moment(TM) and Olive tail moment(OTM),significant difference was found in comets induced by the residual mixture of diflubenzuron in vegetables compared with the control group.This result preliminarily indicated that diflubenzuron itself had no carcinogenicity,but the residual mixture in vegetables could cause mutation and aberration.Therefore,not only its original pesticide,but also its degradation products should be evaluated for their environmental safety.
