桑树悬浮细胞生长规律及其生理特性的研究

摘    要:以桑品种大10为材料,探讨了3种液体培养基B5、N6、MS中的细胞密度变化及悬浮培养细胞的生长规律,并筛选出最适液体培养基。B5液体培养基较适合桑品种大10细胞的悬浮培养;在液体振荡培养条件下,桑树细胞的生长周期为14 d,接种后1~4 d为延迟期,5~10 d为快速生长期,11~14 d为减慢期,14 d以后进入下降期。悬浮细胞分裂指数在快速生长末期达到最大值5.19%,延迟期的细胞活力最强,之后细胞活力开始下降。

Abstract:By using mulberry variety Da 10 as the experimental material,the changes of cell density in 3 kinds of medium B5,N6 and MS,selection of optimum liquid culture medium and the growth regularity of the suspension cells during the growth period were investigated.The results showed that medium B5 was comparatively fit for suspension culture of cells from mulberry variety Da 10;the growth period of mulberry cells was 14 days in the condition of liquid oscillated culture.The lag phase was from the 1st to 4th day,the linear phase was from the 5th to 10th day and the retardation phase was from the 11th to 14th day.The living weight of suspension cells reduced after 14 days.The mitotic index of cell in suspension culture reached 5.19 % in the linear phase.The most powerful viability appeared during the lag phase and thereafter the viability of cell descended.
