雷琼牛Cyt b基因序列分析及其分子系统发生探讨

摘    要:通过对20头雷琼牛(Bos indicus)细胞色素b(Cyt b)基因序列(1 140 bp)的比对和分析,共发现7个变异位点,定义了6种单倍型。经与GenBank上6个牛种的Cyt b基因序列进行比较,分析其碱基组成和核苷酸序列变异,计算不同牛种间的kimura双参数遗传距离。以亚洲水牛(Bubalusbubalis)为外群,应用邻接法构建牛属动物分子系统发育树。结果表明:雷琼牛与印度瘤牛(Bos indicus)一样同属于典型的瘤牛,但两者有明显的区别,推测中国可能是世界瘤牛的发源地之一,不支持雷琼牛含有爪哇牛(Bos javanicus)血统的观点。

Abstract:7 mutation loci and 6 different haplotypes were found when we analyzed 20 cytochrome b(Cyt b) gene complete sequences(1140 bp) of Leiqiong cattles.Through comparing Cyt b complete sequences of Leiqiong cattles with those of 6 bovine species available from GenBank,the base composition and nucleotide variation between these sequences were analyzed and the kimura 2-parameter distance between different bovine species were calculated.Using Bubalus bubalis as outgroup,the phylogenetic tree of genus Bos was constructed by neighbor-joining method.The results demonstrated that Leiqiong cattle belonged to zebu as well as bos indicus,but there were noticeable differences between Leiqiong cattle and Bos indicus,and one of the origin places of zebu maybe distribute in China.Our study didn't support the opinion that Leiqiong cattle contained blood of Bos javanicus.
