摘    要:建立一个测定小麦(TriticumaestivumL.)子粒赤霉素含量的方法。赤霉素与浓硫酸反应,生成三环芳烃衍生物,在414nm处具有最大吸收值,在一定浓度范围内,吸光值与赤霉素的浓度成正比,符合比尔定律。通过扣除小麦子粒赤霉素提取液在414nm处的本底吸光值,排除其他杂质对赤霉素测定的干扰。小麦子粒整粒提取和磨碎提取赤霉素的效果相似。对小麦子粒样品中的赤霉素提取、净化、浓缩等过程进行了优化,建立了一套简单、快速、准确的测定小麦子粒赤霉素含量的方法。该方法的赤霉酸回收率达到97.1%。与经典的荧光测定法相比,本文建立的方法更加简便。利用该方法对几个小麦品种子粒赤霉素含量的测定结果表明,不同小麦品种子粒赤霉素含量具有较大的差异。

Abstract:To establish a determination method for wheat(Triticum aestivum L.) grain gibberellins(GAs).Gibberellins can react with high concentration H2SO4 and form 3-cycle aromatic hydrocarbon derivatives.The derivatives have a top light absorption at 414 nm.In a definite concentration range,the light absorption of the derivatives is positively related with the concentration of GAs,and it's consistent to Bell law.The ground light absorption of wheat grain gibberellin extraction mix at 414 nm was deducted to eliminate the interference of the impurities.The effects on gibberellin extraction with complete grain or crushed grain were similar.The processes of gibberellin extraction,purification and concentration for wheat grain were optimized.A simple,rapid and exact method for wheat grain gibberellin determination was established.The GA3 recovery rate reached 97.1% with this method.This method is simpler and easier to operate than classical fluorescence method.Grain GAs' contents of several wheat cultivars were determined with this method.The result showed that the GAs' contents were significantly different among wheat cultivars.
