几类同核异质雄性不育小麦线粒体DNA的RAPD标记多态性

摘    要:为了明确不同细胞质来源的小麦不育系mtDNA的差异,对具有相同核背景而细胞质分别来源于偏凸山羊草(Ae.ventricosa)、易变山羊草(Ae.Variabilis)、提莫菲维小麦(T.timopheevi)的 3 种小麦细胞质雄性不育系,采用 50条随机引物对其细胞质线粒体DNA(mtDNA)进行了RAPD 多态性分析,筛选出3条特异性引物,其中引物S22 在ms(Ae.ventricosa)90110中扩增出分子量约为1 500 bp 的特异带,引物OPAA16在ms(T.timopheevi)90110中扩增出分子量约为1 200 bp的特异带,引物OPA05在ms(Ae.variabilis)90110中扩增出分子量约为670 bp的特异条带。分析表明,3类不同细胞质的小麦不育系在线粒体DNA上表现出稳定的差异,为进一步在分子基础上鉴别这3类不育胞质源不育系奠定了基础。

Abstract:The mitochondrial DNAs (mtDNA) were analyzed in three wheat male sterile lines with isonucleur and allocytoplasm by RAPD (Random Amplified Polymorphic DNA) technique. 3 specific primers used in the study were screened from 50 random primers. Specific amplified fragment (mt) S22 1500bp, OPAA16 1200bp and OPA05 670bp of CMS in mitochondria DNA were obtained from male sterile lines ms (Ae.ventricosa) 90 110, ms(T.timopheevi) 90 110 and ms (Ae.variabilis) 90 110respectively. The results revealed polymorphism in mtDNA of three wheat male sterile lines, which laid the foundation and provided the theoretical and technical support for further identifying three allocytoplasm male sterile lines in the molecular level.
