摘    要:目的]为花生的转基因研究和白芦藜醇的生物技术生产奠定基础。方法]利用发根农杆菌R1601分别侵染花生的子叶、叶片和茎段,诱导毛状根;利用PCR技术检测毛状根。结果]叶片和茎段在发根农杆菌R1601侵染后产生毛状根,但叶片诱导率较高,其诱导率达65.6%,更适合作为转化外植体。毛状根除菌后,能在MS培养基上自主生长。PCR扩增表明,T-DNA序列整合进花生的基因组中。结论]建立了发根农杆菌介导的花生遗传转化体系。

Establishment of Peanut Genetic Transformation System by Agrobacterium rhizogenes
Abstract:Objective] The aim was to lay a foundation for the research on transgenic peanuts and the biotechnology production of resveratrol.Method] A.rhizogenes R1601 was used to infect the cotyledon,leaf and stem of peanut and to induct hairy roots.PCR technology was applied to detect hairy roots.Result] Hairy roots were produced after the leaf and stem infection of A.rhizogenes R1601,with leaf being higher induction rate(65.6%),and it was more suitable as transformed explants.After eliminating bacteria with antibiotic,the hairy roots could grow independently on MS medium.PCR identification showed that T-DNA sequence was integrated into peanut genome.Conclusion] The A.rhizogenes-mediated genetic transformation system of peanut was established.
