摘    要:[目的]研究Alcalase蛋白酶对大豆分离蛋白的水解作用及水解物的性质。[方法]通过单因素试验,研究pH值、温度、酶浓度、底物浓度等因素对Alcalase蛋白酶酶解大豆分离蛋白的影响,通过正交试验确定Alcalase蛋白酶水解大豆分离蛋白的最佳水解条件。[结果]Alcalase蛋白酶水解大豆分离蛋白的最佳水解条件是pH值8.0、温度60℃、酶浓度1000U/g、底物浓度3%,水解时间2h,大豆分离蛋白水解度为46.13%。[结论]酶解后大豆分离蛋白的水解度达到了制备大豆多肽的要求。

Abstract:[Objective] This paper seeks to study hydrolysis process for soybeen protein isolate with Alcalase and the nature of the hydrolyzate was studied. [Method] The effects of the main parameters including pH,hydrolysis temperature,enzyme concentration,substrate concentration were investigated. The optimum conditions had been found by single-factor and orthogonal tests. [Result] The optimum conditions were showed as following:pH was 8.0,hydrolysing temperature was 60 ℃,enzyme concentration of substrate was 1 000 u/g,substrate concentration was 3%,hydrolysis time was 2 h,degree of hydrolysis was 46.13%.[Conclusion]After enzymatic hydrolysis,the degree of SPI reached preparation of soybean peptides requirements.
