摘    要:锌指蛋白核酸酶(Zinc Finger Nuclease,ZFN)是一种人工合成酶,含有锌指蛋白和FokⅠ核酸内切酶的剪切结构域,因其能特异性识别并切割DNA序列及其可设计性而被用基因定点突变和外源基因定点整合。这项技术已应用于转基因动物研究中。本文综述了锌指核酸酶技术在转基因研究中的应用进展,并对锌指核酸酶技术的应用前景进行了展望。

Abstract:Zinc finger nuclease(ZFN) is a kind of artificial synthesis containing zinc finger protein and the shear structure domain of FokⅠ endonuclease.Because it can specifically identify and cleave DNA sequences and easily be designed,it is used for the site-directed genetic mutations and integration of exogenous genes.This technology has been applied to the research of transgenic animals.In this paper,the research progress of zinc finger nuclease technology and its application were reviewed,and its application prospect was also forecasted.
