核果类果树基因组DNA提取方法的研究

摘    要:以核果类果树油桃、扁桃、杏、山桃、榆叶梅和樱桃的幼叶为试材,进行了基因组DNA的提取方法的比较研究,其中改良SDS法效果最好;采用改良SDS法提取油桃的幼叶、花蕾和韧皮部基因组DNA的效果表明,三者均可以作为基因组DNA提取的材料,且以幼叶效果最理想;采用改良的SDS法可在自然阴干的扁桃和油桃叶片中提取到完整的DNA;将改良SDS法提取的油桃不同部位和自然阴干的扁桃和油桃叶片基因组DNA,经SSR-PCR检测发现,扩增效果清晰稳定,完全可以应用于果树分子生物学研究.

Abstract:Drupe fruit trees contain plenty of polysaecharides,polyphenels,pigments,tannin and other secondary metabolites,which affects the genomic DNA extraction.In this research,the genomic DNA was extracted from the fresh young leaves of some drupe trees,the results showed that the improved SDS method was the best for DNA extraction.The improved SDS method was applied to extract genomic DNA from different organs of nectarine,the results showed that young leaves,flowers and basts could be used to extract the genomic DNA、Besides,the genomic DNA could also be extracted from the leaves of the almond and nectarine which were dried in the shade by improved SDS method.Through the SSR-PCR detection,the extracted DNA had a good qulity for modecular biological study.
