阿月浑子性别鉴定的RAPD分析

摘    要:应用随机扩增多态性DNA(RAPD)技术对新疆阿月浑子(PistaciaveraL.)地方品种雌、雄株进行性别鉴定的研究。通过叶片总DNA抽提、RAPD标记分析及反复的试验筛选,找到了一个适用于新疆阿月浑子的RAPD反应体系和循环参数。采用OPO08引物对雌、雄株基因组DNA扩增出性别之间差异性的核苷酸片段,证实雄性植株DNA的扩增产物有一条大约700bp的特异性条带,而雌性植株则无此特异性条带。即找到一条与阿月浑子性别相关的基因标记,表明RAPD技术可应用于新疆阿月浑子地方品种的性别鉴定。此研究是对我国雌雄异株果树阿月浑子在分子水平进行早期性别鉴定的一个尝试。

Abstract:The sex identification of pistachio(Pistacia vera L.)cultivars in Xinjiang were studied by RAPD(Random Ampli-fied Polymorphic DNA)analysis.The reaction systems and cycling parameters of RAPD for cultivars in Xinjiang were con-firmed after the reduplicate experiments of genomic DNA abstraction and RAPD analysis.The results showed that a special oxyribonucleic acid band of about 700bp was obtained from the male plant by using primer OPO 08 to amplify the genomic DNA abstracted from the leaves of male and female trees,indicating that a gene mark related to sex of pistachio was found which could be used for the sex identification of pistachio cultivars in Xinjiang.This research was an attempt to identify the sex in early stage of the tree of Pistacia vera L.on molecular level in China.
