GFP与GnRH/TRS融合基因表达载体的构建及表达

摘    要:应用基因工程技术构建GnRH/TRS与绿色荧光蛋白的融合基因,核酸序列测定和Western boltting印迹分析基因表达;激光共聚焦荧光显微镜观察活细胞内荧光布局。结果融合基因获得了正确表达,GnRH/TRS-GFP融合基因的瞬间和稳定表达均获得了相同结果。GnRH/TRS-GFP融合蛋白具有绿色荧光蛋白的自发荧光特性,且不影响Gn-RH/TRS分子在细胞内的正确表达,为低促性腺激素性功能减退综合征治疗的进一步研究奠定了基础。

Abstract:A fusion gene composed of green fluorescent protein(GFP) and GnRH/TRS DNA was constructed by standard cloning technology.Nucleotid sequence and Western blot analysis were used to evaluate the expression of the fused gene.Laser-scanning confocal microscopy was used to monitor the distribution of green fluorescence.Laser-scanning confocal microscopy clearly revealed that GnRH/TRS-GFP could be transiently and stably expressed in mammalian cells.The GFP tag did not interfere with the natural assembly and transport of GnRH/TRS molecule,and GnRH/TRS-GFP could act as a potential tool to learn more about GnRH/TRS molecules in living cells for further foundations established in the scientific research and the real application of treat of Gonadotropin-releasing hormone difficiency in the human(idiopathic hypogonadotropic hypogonadism and kallman's syndrom).
