摘    要:本研究利用ISSR分子标记技术对12个优良观赏枫香种质资源进行遗传多样性研究。从12份不同枫香单株叶片样本中提取DNA,设计适应枫香的ISSR扩增正交体系优化实验,筛选出19条ISSR引物得到127条扩增带,其中多态性条带为94条,多态性条带比率为74.01%。再利用ISSR分子标记技术对数据进行遗传多样性和相似度分析,发现除4号和11号样本具有较高的遗传相似度0.83外,其他样本的遗传相似度为0.61~0.79,清楚的显示其优良种质的遗传差异性,具有较高的遗传多样性。

Abstract:The research did research on genetic diversity of 12 Liquidambar formosana which has good ornamental germplasm on the basis of ISSR analysis.From 12 different leaf samples extracted DNA,the authors designed an orthogonal optimization experiment that was suitable for propagating ISSA of germplasm.In the experiment,we selected out 127 amplified bands from 19 ISSR primers,of which 94 bands(74.01%)were polymorphic.Then we used the data to analyze the genetic diversity and similarity on the basis of ISSR analysis.The results showed that the genetic similarity of samples of No.4 and No.11 was 0.83,and other samples' genetic similarity differed from 0.61 to 0.79.So it was clearly concluded that good varieties had genetic differences and genetic diversity.
