| 摘 要： | 本试验以盆栽三年生辽伏/海棠为试材,研究了苹果新根细胞分裂素的提取、纯化和高效液相色谱(HPLC)分析方法。苹果新根用80%冷甲醇抽提;在pH2.5下用石油醚和pH8.0～8.5下用正丁醇萃取,然后用pH3.5磷酸缓冲液做流动相的PVP柱层析和sep—Pak C_(28)筒柱进行纯化;最后用甲醇、水溶剂系统(pH3.5)在ZORBAX ODS柱上用离子对色谱法进行分离、鉴定。苹果新根中的CTK种类主要有Z和ZR.

| Abstract: | Abstraction and purification of endogenous cytokinins in new apple roots and their efficiently analytical methods of liquid chromatography were studied. Three-year-old pot-cultured apple ( Var.Liao-fu/M. Prunifolia)was used as material for the experiment. The results were as follows: 1.Cytokinins were extracted from roots with 80% cold methanol, initially purified with petroleaum and n-BuoH, followed by chromatography on insoluble polyvinylpyrrolidone (PVP) and Sep-pak C_(18) cartidges, and finally separated and quantified with mothanol-water on ZORBAX ODS reversed phase HPLC columns. The experimental conditions were compared in every step of analytic procedure according to the characters of the experimental materials. 2.Cytokinins in apple roots had two typcs: both zeatin and zeatin riboside. |
