马铃薯耐热无性系离体筛选的初步研究

摘    要:为了获得耐热性较高的马铃薯品种,本研究以马铃薯试管苗的愈伤组织为供试材料,采用羟脯氨酸(HYP)和热胁迫相结合的方法来进行马铃薯耐热无性系的离体筛选研究.研究结果表明,MS+1.5~3.0 mg/L6-BA+0.1 mg/L NAA的培养基均能够诱导全部供试马铃薯茎段产生愈伤组织,说明细胞分裂素6-BA对愈伤组织的形成有着非常重要的作用;同时确定了马铃薯愈伤组织耐羟脯氨酸筛选的适宜浓度为0.2~0.4 mg/mL.MS+1.0~5.0mg/L BA+0.1 mg/LNAA+5 mg/LGA的分化培养基能够诱导愈伤组织进行植株再生,但分化率较低,只有8%~25%.结果表明不同浓度的分化培养基均能诱导马铃薯愈伤组织分化成苗,本实验还筛选出热胁迫的适宜条件为42℃24 h,且经过羟脯氨酸与高温胁迫结合多次筛选获得了两个耐热无性系NKY1和NKY2.初步表明本筛选方法可以用于马岭铃薯耐热无性系的筛选.

Abstract:In order to obtain the heat-tolerance potato cultivars, we employed the potato test-tube plantlet callus as materials for the selection of heat-tolerant clonal lines of potato in vitro by using the combinative method of Hydroxyproline-resisitant and high temperature stress in this research. The results showed that all the tested potato stem segments can be induced to generate callus by the medium MS+ 1.5~3.0 mg/L 6-BA+0.1 mg/L NAA, which illustrated that the cytokinin 6-BA played an very important part in the callus formation. Meanwhile, we have also found that the optimal concentration of HYP for selection is 0.2~0.4 mg/mL. The differential medium MS+ 1.0~5.0 mg/L BA+0.1 mg/L NAA+5 mg/L GA can induce the callus for plantlet regeneration, but the differentiation rate was comparatively low, only between 8% and 25%. Experiments revealed that differential medium of different concentrations can induce the patato callus differentiate into seedling. We have determined that the optimal condition of high temperature stress selection was 42℃ for 24 hours. After repeated tests, we acquired two heat-tolerant potato clonal lines, designated as NKY1 and NKY2. Preliminary data show that this kind of selection approach is suitable for the selection of heat-tolerant potato clonal lines.
