菜豆几丁质酶基因转化小麦的研究

摘    要:利用农杆菌二元载体转化法和花粉管通道法 ,以菜豆几丁质酶基因转化小麦东农 774 2。农杆菌感染后的愈伤组织以 G4 183 0 mg· L-1筛选 5周 ,经 PCR和 PCR- Southern杂交检测 ,有0 .3 8%的愈伤获得转化。花粉管通道法转化小麦共操作小花 3 85朵 ,其中有 68.3 0 %的小花结实 ,经 PCR和 PCR- Southern杂交检测 ,获得转基因植株的转化频率为 0 .52 %。

Abstract:Bean chitionase gene was transformed into spring wheat Culfivar Dongnong 7742 by Agrobacterium tumefacicns-mediated transformation and pollen tube pathway.The callia infected with Agrobacterium tumefaciens has been selected for five weeks by G418 30mg/L,and tested by PCRand PCR-southern Blotting.The results indicated that 0.38% callia has been transformed successfully.However,385 little buds have been operated using pollen tube pathway,then 68.30% buds breeded.The frepuency of transgenic plants was 0.52% by PCR and PCR-suthern Blotting testing.
