| 摘 要： | 以富含类黄酮的月季花瓣为试验材料，通过比较不同方法，优化和改进提取步骤，发现CTAB多级沉淀法、改良异硫氰酸胍法提取的RNA的完整性和纯度较好，电泳显示28S和18S条带清晰，28S条带亮度是18S的两倍。 而SDS-LiCl法、RNArose Reagent试剂快速法提取的RNA黄酮胶状沉淀难溶，电泳图谱28S和18S条带模糊难辨，5S条带很亮。这为月季花色差减文库构建及花色相关基因克隆奠定了良好的基础。

| Abstract: | Diverse,inexpensive,and reliable protocol for the extraction of RNA from Rosa hybrida corolla tissue containing large quantities of polyphenols,procyanidins,and poly-saccharides are compared.Results show CTAB-many-level-precipitation method and improved isothiocyanate method are efficient for isolating RNA with higher purity and integrality while RNArose Reagent method and SDS-LiCl method are not.These provide convenient and effective methods for research on gene expression in anthocyanin biosynthesis and change in flower color. |
