摘    要:研究了离子霉素处理时间、处理浓度和不同胚胎基础培养基对猪无透明带卵孤雌激活的影响。结果表明:(1)10 μM离子霉素处理5 min、7 min、10 min的激活率64.81%、66.67%、68.75%差异不显著(P>0.05),但显著高于处理3 min的激活率36.17%。(2)分别以10 μM、15 μM离子霉素处理10 min的激活率68.18%、65.11%差异不显著(P>0.05),但显著高于5 μM的激活率40.54%(P<0.05)。(3)分别以NCSU-23+4%BSA和TCM199+4%BSA作为基础培养基进行培养,其激活率68.51%、62.50%无显著差异(P>0.05)。试验结果表明:10 μM离子霉素处理5 min联合2 mmol/L6-DMAP处理2 h激活, 以NCSU-23+4%BSA为胚胎基础培养基能有效提高猪无透明带卵孤雌激活的激活率。

Abstract:The effects of ionomycin dealing time, ionomycin dealing density, different basic embryo culture mediums on parthenogenetic activation of porcine zona-free oocytes were studied. The results are as follows: (1) The activation rate with 10 μM ionomycin dealing 5 min(64.81%) was not significantly different from that of dealing 7 min(66.67%), 10 min(68.75%), but significantly higher than 3 min(36.17%). (2) The activation rate of ionomycin dealing 10 min with 10μM(68.18%) was not significantly different from that with 15μM (65.11%), but significantly higher than that with 5μM (40.54%). (3) The activation rate of using NCSU-23+4%BSA and TCM-199+4%BSA as embryo basic culture mediums was not significantly different(68.51%, 62.50%). The results of this experiment showed that 10 μM ionomycin dealing 5 min in combination with using NCSU-23+4%BSA can effectively activate porcine zona-free oocytes.
