影响小麦RAPD稳定性的试验技术研究

摘    要:随着PCR和PAPD技术在生物学业领域的广泛应用,人们在实验中遇到的问题也越来越多,其中PAPD的可重复性差是最主要的一个问题,也是该技术最难克服的特点。作者在利用RAPD技术标记进行小麦的抗病毒基因研究中,对影响扩增结果的模板、底物、引物和扩增程序等进行了实验探索,结果表明:模板的浓度、dNRP浓度、引物浓度及反应程序均对RAPD扩增结果有明显影响。

Abstract:With the wide application of RAPD (Random amplified polymorphic DNA) in biology, more and more problems were presented in our research. All of these problems, repeat ability was the most difficult one to overcome. For this reason the authors tried to probe into some essential factors which might influence the results from the research of tagging the resistance gene to the wheat virus with RAPD technique. These factors that include templet, raction substrate, primer and procedures of RAPD were studied in this paper. The experiment results showed that concentration of the templet, dNTP, primer and procedures had apparent influence on the RAPD test.
